The rapid antigen test looks for the presence of Sars-Cov-2 virus proteins in the sample and requires the same nasopharyngeal swab as the PCR test but involves a simplified analysis technique. The Covid-19 rapid antigen test consists of a manual test, with direct reading on a small plastic box. The result is displayed in less than 15 minutes in the form of bars that appear on the box. Its advantage lies in the speed of rendering of the result. The antigen test is useful for a person who has symptoms related to Covid-19 (cough, fever), because it allows you to obtain a result quickly.

The COVID-19 Antigen Test Kit (Side Chromatography) is designed to detect the presence or absence of SARS-COV-2 nucleocapsid proteins in respiratory specimens from patients with signs and symptoms of suspected infection of COVID-19. When samples are processed and added to the test device, the SARS-COV-2 antigens present in the sample bind to the colloidal gold-labeled antibodies in the test strip. Antigen-antibody complexes migrate through the test strip to the reaction zone and are captured by a line of membrane-bound antibodies.
- A positive result is determined when antigen-antibody complexes are deposited in the Test "T" position and the Control "C" position on the assay device to form colored lines.
- A negative result is determined when only the " C" position of the control forms a colored line, but no visible colored line appears on the " T" position of the test.
- For quality control to ensure the validity of the test result, a colored line must always appear on the "C" position of the control, regardless of whether the result for the "T" position of the test is positive or negative. If there is no visible line at the "C" position of the control, the test kit result is invalid and a new test should be performed.
Materials Provided - Each box of test kit contains:

Material required, but not included
- Timer or watch
- Personal protective equipment, such as protective gloves, medical mask, goggles and lab coat.
- Cleaning containers and appropriate disinfectants for hazardous waste.
- 1.0ml calibrated micropipette with pipette tips.
Nasal swab sample collection;
- To collect a nasal swab sample, carefully insert the swab (provided in the kit) into the nostril that has the most secretions on visual inspection. Using a slight rotation, push the swab until resistance is met at the turbinates (approximately 2.0 – 2.5 cm). Roll the swab about 5 times against the nasal wall and then remove it from the nostril.
Sample preparation;
- Unscrew the cap from the Sample Extraction Buffer bottle, squeeze the Sample Extraction Buffer bottle, and add the Sample Extraction Buffer to the Extraction Tube (Figure 2). After sample collection is complete, immediately immerse the swab in the Sample Extraction Buffer. (Illustration3) At the end, ensure that the extracted sample is completely soaked in the swab buffer, rotate the swab against the wall of the tube repeatedly for 10 seconds, then use the fingers to hold the tube (tube wall is semi-solid). Squeeze the swab several times while slowly withdrawing it from the tube. (Illustration 4) The purpose of the swab squeezing against the wall of the tube is important, because it can keep the sample containing liquid in the tube as much as possible. After removing the swab and disposing of it in a special waste container, place it in a nozzle (provided in the kit). Screw the top of the extraction tube tightly and shake the tube gently to mix the liquid inside well. (Picture5)
Interpretation of results
- Positive Result: The Test "T" position and the Control "C" position both appear with visible colored lines. If the “ T ” position of the test shows a pale color, the result is also positive.
- Negative result: only the control position " C" appears in color, while the test position " T" is empty.
- Invalid result: if the " C " control remains empty without the color appearing, the test result is invalid, because the detection function did not work. It is therefore necessary to retest the sample.
